چکیده

Despite enormous therapeutic advancements, the incidence of hypertension continues to rise and remains a medical burden of global proportions. This is likely due, in part, to the complex pathogenesis of hypertension. In addition to pathological processes in peripheral cardiovascular organs, the central nervous system (CNS) has emerged as a major culprit. The renin–angiotensin system (RAS) plays a crucial role in cardiovascular control, and both systemic angiotensin-II (Ang-II) and centrally generated Ang-II signal through complex neural networks to contribute to the development of hypertension (Figure). Although the effects of Ang-II within the brain are multifaceted, several key hypertensive players have emerged, including oxidative stress, endoplasmic reticulum stress, inflammation, and transcription factor activation. The purpose of this review is to provide a brief update on recent work related to Ang-II, the brain, and hypertension with a specific focus on Hypertension articles when appropriate.

Crocin prevents acute angiotensin II-induced hypertension in anesthetized rats

Objective: Angiotensin II (Ang II), the main product of renin-angiotensin system (RAS) has a well-known role in cardiovascular regulation. Over-production of Ang II is one of the important underlying mechanisms of hypertension. In this study, the effect of crocin on cardiovascular responses in rats with acute hypertension induced by Ang II was evaluated. Materials and methods: Rats were divided...

Intrarenal angiotensin II augmentation in angiotensin II dependent hypertension.

In several models of angiotensin II (ANG II) dependent hypertension, intrarenal ANG II levels increase to a much greater extent than the circulating levels even though the renal renin levels are decreased. The 2-kidney-1-clip (2K1C) Goldblatt rat model is particularly intriguing because hypertension develops in the presence of an intact kidney which would be expected to maintain sodium balance ...

Renal uptake of circulating angiotensin II in Val5-angiotensin II infused rats is mediated by AT1 receptor.

Previous studies have demonstrated that augmentation of intrarenal angiotensin II (ANG II) levels during ANG II induced hypertension involves both endogenous formation and accumulation of circulating ANG II. The present work extends these findings and determines whether accumulation of infused ANG II in the kidney requires AT1 receptor activation by using Val5-ANG II as the infused peptide. Mal...

Regulation of intrarenal angiotensin II in hypertension.

Intrarenal angiotensin II (Ang II) is regulated by several complex processes involving formation from both systemically delivered and intrarenally formed substrate, as well as receptor-mediated internalization. There is substantial compartmentalization of intrarenal Ang II, with levels in the renal interstitial fluid and in proximal tubule fluid being much greater than can be explained from the...

Role of brain corticosterone and aldosterone in central angiotensin II-induced hypertension.

Circulating angiotensin II (Ang II) activates a central aldosterone-mineralocorticoid receptor neuromodulatory pathway, which mediates most of the Ang II-induced hypertension. This study examined whether specific central infusion of Ang II also activates this central aldosterone-mineralocorticoid receptor pathway. Intracerebroventricular infusion of Ang II at 1.0, 2.5, and 12.5 ng/min for 2 wee...
